Supplementary Figure 1: Synchronised HeLa cells, either untreated (Con) or treated with 100 μg/ml SBHA (SBHA)
were harvested as cells progressed into mitosis (M) or in the subsequent G1 phase (G1).
Supplementary Figure 2: HeLa cells were either fixed (Con), or permeablised then fixed (Perm) and stained for HP1β, the centromeric marker ACA and DNA.
Supplementary Figure 3: Asynchronously growing MCF7 cells were either untreated or treated with 100 μg/ml SBHA for 24 h, fixed without permeablising first and stained as in D.
Supplementary Figure 4: HeLa cells were synchronised and either untreated (Con) or treated with 100 μg/ml SBHA in S phase (HDACi). Cells were harvested 24 h after synchrony release, permeablised and fixed then stained for HP1β, ACA and DNA.
Supplementary Figure 5: (A, B) HeLa cells were transfected with either Jmjd2C or an inactive mutant (DD). 24h after transfection, cells were fixed and stained for di-methyl (2Me) or tri-methyl (3Me) H3K9, and DNA. Jmjd2C expression was marked by GFP.

HeLa cells treated with 100 μg/ml SBHA for 6 h were used as a control. (B) HeLa cells, either untreated (Con) or treated with 100 �g/ml SBHA for either 10 or 24 h were immunoblotted for H3K4 3Me or total H3.
