The RGS2 gene product from a candidate hypertension allele shows decreased plasma membrane association and inhibition of Gq

Data Supplement

Files in this Data Supplement:

  • Data Supplement - Supplemental Figure 1. Non-corrected spectra of RGS2 and R44H peptides. RGS2 helix domain peptides corresponding to residues 34-57 from WT and R44H were incubated with increasing amounts of purified unilaminar liposomes from brain lipid extracts. Tryptophan fluorescence emission spectra of W41 were collected as described. Control spectra were collected as described above except no protein was added.
  • Data Supplement - Supplemental Figure 2. TFE results in helix formation of both WT and R44H mutants without affecting tryptophan emission maxima. RGS2 WT and R44H peptides were incubated in either PBS or PBS containing 15% TFE. Tryptophan emission spectra and CD spectra were collected as described previously after a 5 minute incubation period. Shown are corrected spectra where background was subtracted from the protein spectra.
  • Data Supplement - Supplemental Figure 3. Non-corrected CD spectra of RGS2 and R44H peptides. CD spectra were obtained as described above for both RGS2 and R44H peptides in either PBS or lipid solution. Shown are non-corrected peptide spectra and the corresponding control spectra.
