Purified sGC (0.1 μg of sGC) was preincubated for 3 min at 37° with 1 μm DEA-NO and the indicated YC-1 concentrations in the absence of substrate. After 3 min, sGC was diluted 5-fold with incubation buffer containing substrate and YC-1, yielding the indicated final concentrations. sGC activity was then determined during 10 min at 37°.
