Effects of squalestatin 1 treatment on sterol biosynthesis in primary cultured rat hepatocytes

The 48-hour primary cultured rat hepatocytes were treated for 24 hr with medium alone or containing 10−8 or 10−7 m squalestatin 1. At 2 hr before harvest at 72 hr, culture medium was supplemented with 10 μCi of [14C]acetate. Nonsaponifiable lipids were extracted from the cells and separated by thin layer chromatography, and amounts of radioactivity incorporated into bands comigrating with authentic samples of cholesterol, lanosterol, or squalene were measured.

    • Results are presented as mean ± standard deviation (three dishes/treatment group), except for the untreated cholesterol value, which is presented as mean ± range (two dishes).
