One unit corresponds to 1 pmol of dNTP incorporated into acid-insoluble materials over 60 min by using as substrate an activated calf thymus DNA preincubated with DNAse I. The relative replication activity is the amount of replication products normalized by the amount obtained without either excess Pol β or ddC. Quantification analysis of the resolved radioactive bands on the gel was achieved with the use of PhosphorImager Storm system analysis and ImageQuant software.
