The binding of agonist and antagonist ligands to α1A-1-adrenoceptor-Gα11 fusion proteins
Fusion proteins incorporating both wild-type and point mutant forms of the human α1A-1-adrenoceptor and Gα11 that result in a lack of information transfer from receptor to G protein in response to agonist ligands were expressed transiently in HEK293 cells and membranes prepared. The binding affinity of [3H]prazosin and both phenylephrine and adrenaline was then estimated from ligand binding studies.
