DNA-protein cross-linking in cells

DNA-protein CL formation following exposure of A2780 and A2780cisR cells for 48 h to cisplatin or adamplatin(IV). After washing, cells were subjected to the potassium-SDS procedure following overnight storage at −70°C. The cross-linking was evaluated using the DPC defined as a ratio of the amount of SDS-precipitable DNA in treated cells to the amount of SDS-precipitable DNA in control cells (Zhitkovich and Costa, 1992). The values of DPC shown are those normalized to the identical amount of platinum associated with the cells. Each value of DPC represents the average of four samples ± S.E.
