TABLE 2 Mutational effect on glutamate-induced Ca2+-response and cell surface expression of mGluR5
BHK cells were transiently transfected with wild-type and mutated mGluR5 using the calcium phosphate method. The intracellular Ca2+ response was measured on an imaging-based plate reader. Cell surface expression of wild-type and mutated receptors was determined by cell surface ELISA. Results are presented as mean ± S.E.M. and represent data from minimum two independent transfections. n states the number of experiments (duplicate measurements). Mutants were compared with wild-type mGluR5 (Emax, pEC50, and OD) by one-way ANOVA combined with Dunnett's multiple comparison test.
