Regulation of nitric oxide/reactive oxygen species redox signaling by nNOS splicing variants
2022, Nitric Oxide - Biology and ChemistryCitation Excerpt :Therefore, the absence of BH4 results in monomerization and, thereby, in nNOS uncoupling to produce more ROS, resulting in cytotoxicity. The formation of nNOS translational splice variants is also thought to be one of the important processes regulating the nNOS function [23–25]; the principal isoform expressed in the brain is nNOS-α. Although nNOS-μ was first detected in skeletal muscles and the heart [25], we later identified the expression of nNOS-μ in the rat brain, which differs from that of nNOS-α, especially in the cerebellum [26].
Variants of Neural Nitric Oxide Synthase in the Spinal Cord of Neuropathic Rats and Their Effects on Nuclear Factor-κB (NF-κB) Activity in PC12 Cells
2009, Journal of PainCitation Excerpt :However, 3 splice variants were confirmed by Southern blot hybridization with a rat nNOS exon3-specific and [γ32] P end-labeled oligonucleotide. These 3 variants corresponded to nNOSα-a (exon1a/exon2), nNOSα-b (exon1b/exon2), and nNOSβ (exon1a/exon3), respectively, according to published data.11,14,20,27,31 Translation of the nNOSα-a and nNOSα-b would result in the one protein, nNOSα, and the nNOSβ mRNA, with exon2 spliced, could be translated into the nNOSβ protein, which lacks the PDZ binding domain encoded by exon2.
Stimulation of the cAMP pathway protects cultured cerebellar granule neurons against alcohol-induced cell death by activating the neuronal nitric oxide synthase (nNOS) gene
2007, Brain ResearchCitation Excerpt :After concentration determination on a UV spectrophotometer (Eppendorf Biophotometer), 250 ng of cDNA was used as the template for amplification of the nNOS message. The real-time quantitative PCR primers and probe for mouse nNOS were designed to cross an intron, using published sequences ((Ogura et al., 1993); GeneBank Access. # NM008712).
