The Dendritic Translocation of Translin Protein in the Form of BC1 RNA Protein Particles in Developing Rat Hippocampal Neurons in Primary Culture

Abstract

Neural BC1 RNA is distributed in neuronal dendrites as ribonucleoprotein particles (RNP). Our previous studies indicated the presence of Translin in BC1 RNPs, which is a translational repressor and links a subset of mRNAs to microtubules. In this study, we confirmed that Translin associates with BC1 RNP and we used immunocytochemical methods to examine the subcellular distribution of Translin in developing hippocampal cells in primary cultures. Translin was detected in both the nuclei and cytoplasm of neurons, whereas in glial cells it was localized in the nuclei. Consistent with the reported developmental time course of BC1 RNA expression and dendritic delivery the translocation of Translin to the neuronal dendrites appeared to correlate with neuronal development and differentiation events such as the onset of synaptogenesis in culture. These observations suggest that BC1 RNP or Translin itself may be relevant to the dendritic translation of mRNAs in response to transsynaptic activity.

      Similarly, the immunoprecipitation of KIF5 has identified synaptic transcriptomes that are transported to neuronal dendrites in Aplysia neurons (Puthanveettil et al., 2013). Consistent with these reports, dendritic transport of BC1 lncRNA occurs in association with dendritic RBPs, such as Staufen1/2, Translin and FMRP (Kobayashi et al., 1998; Mallardo et al., 2003; Zalfa et al., 2003). These observations suggest that a mapping of lncRNA-RBP interaction is imperative to bolster our understanding of targeted trafficking of lncRNAs to translationally competent compartments.

      TRAX and translin are present at high levels in neurons. Mutagenesis studies show that these proteins play key roles in function and development in cultured cells of neuronal linage and in the neural system of whole animals, although the underlying basis is unknown (Finkenstadt et al., 2000; Kobayashi et al., 1998; Stein et al., 2006; Wu et al., 1997).       BC1 contains a specific cis-acting element that is required for its transport to dendritic processes in an activity-dependent manner (Muslimov et al., 1998, 1997). Though the transport RNP that mediates the dendritic sorting of BC1 is not fully characterized, it likely contains translin and staufen proteins based on biochemical fractionation experiments (Kobayashi et al., 1998; Mallardo et al., 2003). BC1 was implicated in the regulation of translation in dendrites (Wang et al., 2002), evidently in association with the fragile X syndrome protein FMRP (Zalfa et al., 2003), though this finding remains controversial (Iacoangeli et al., 2008; Wang et al., 2005).

      Indeed, translin has been shown to regulate dendritic targeting of BDNF mRNA through a sequence within the coding region of BDNF (Chiaruttini et al., 2009). Translin is associated with regulation of local protein synthesis as well as learning and memory (Kobayashi et al., 1998; Muramatsu et al., 1998; Finkenstadt et al., 2000; Li et al., 2008), and translin knockout mice have altered spatial memory, fear learning, and anxiety behaviors (Stein et al., 2006). These data suggest that altered local BDNF production could contribute to synaptic dysfunction in patients and warrant further investigations into the connection between altered BDNF synthesis at synapses and psychiatric disorders.
