In our hands, Fe(DETC)2 – which is normally used as a spin trap in EPR experiments [49] – works reasonably well as NO scavenger as the latter is incorporated into an adduct rather than released as reactive, higher nitrogen oxide [48]. If DAF-fluorescence is compared between two conditions, one of them characterized by increased in reactive species, like inflammation/no inflammation, shear stress/static conditions, or normoxia/hypoxia, further controls are needed.

The formation or presence of fluorescence signals unrelated to NO (adducts with, e.g., ascorbate or dehydroascorbate [27]) can be excluded by applying analytical separation techniques.
