On the other hand, RhoA has been widely implicated in cellular responses to mechanical stress [98,99], and plays a critical role in assembling actin stress fibers in response to various stimuli [100–102], which are mediated in part by one of its downstream effectors, the Rho-associated protein kinase (ROCK). Cells grown on stiff matrices assemble robust actin stress fibers and mature focal adhesions, in contrast to the poorly defined, disorganized F-actin network and small focal adhesions in cells on soft matrices [103], suggesting that RhoA activity increases with increasing ECM rigidity.
