Abstract

A delta opioid receptor complementary DNA (cDNA) was cloned by expression of cDNA library from activated thymocytes in Cos 7 cells. The deduced amino acid sequence of this receptor was similar to that described in the brain. As analyzed by southern blot hybridization, the delta opioid receptor transcripts are constitutively expressed in unactivated thymocytes. In addition, neither κ nor μ opioid receptor transcripts were detected in thymocytes, suggesting tissue-specific opioid receptor gene expression in the immune system. The studies represent the first report of a full-length opioid receptor in the immune system.

In fish, C. punctatus, β-end differentially regulates the phagocytic and cytotoxic activities of splenic phagocytes. It stimulated the phagocytosis and respiratory burst activity, while inhibited the nitric oxide production, and thus, helps in maintaining immune homeostasis [54]. However, in most of the studies in mammals, β-end is shown to downregulate the phagocytic [44,63] and proinflammatory activities [52].

Our results are in agreement with previous findings showing that adrenalectomy abolished the apoptotic effect of morphine on thymocytes but not on splenocytes.165152 Moreover, only δ and κ opioid-receptor subclasses have been observed on thymic cells.53-55 Together, our findings indicate that, under normal conditions or in an IgG pool for therapeutic use, anti-hMOR IgG autoantibodies could participate in immunoregulatory processes by favoring Fas-mediated control of the immune response.
