If P2X are always an assembly of three subunits and the ligand binding site is the extra cellular loop between the two TM domains in each subunit, then for the receptor to function as an ion channel, three molecules of ATP are required (Bean, 1990, 1992; Roberts et al., 2006a). If there is a mutation such that ATP does not bind then the receptor would be immediately dysfunctional.
