This chapter discusses the use of tetracycline-controlled gene expression systems to study mammalian cell cycle. Systems that permit the activity of individual genes to be quantitatively controlled are particularly useful for the study of cell cycle progression and arrest. They allow varying the concentration of particular proteins within a cell and to relate the resulting physiological changes to specific gene products. Moreover, they are useful for identifying components that interact with a protein under study, and thus may help to elucidate crucial signaling pathways. This chapter outlines the Tet expression systems, the vectors currently available, and their use in developing stable cell lines that allow conditional expression of a target gene. Because only routine techniques in cell culture and molecular biology are required for applying these control systems, the reader is referred to an earlier report in this series for more technical discussions.
