An intriguing hypothesis is that synaptically released glycine could bind to and activate NR3 subunit-containing NMDARs (Chatterton et al., 2002; Smothers and Woodward, 2007) which are expressed in the adult hippocampus (Bendel et al., 2005). However, IPSCs were completely abolished by strychnine (Fig. 10B) which is ineffective at NR3-containing NMDARs (Smothers and Woodward, 2007), ruling out this hypothesis.

At the cellular level, the expression of GluN3 has been confirmed in several neuronal cell types, including pyramidal neurons of the neocortex and hippocampus [8,31,38,41,42] and cerebellar Purkinje and granule cells [31,38,43]. GluN3A and GluN3B are located in both neuronal somata and neurites [31,38,42], and they co-assemble with other NMDAR subunits in the postsynaptic density of asymmetric synapses [18,31,32,44,45].
