TRPV1 involvement in inflammatory tissue fibrosis in mice
2011, American Journal of PathologyCitation Excerpt :These results further indicate that injury-induced TRPV1 activation on resident stromal cells rather than on infiltrating inflammatory cells determines the outcome of the wound healing response. Similar findings of suppression of tissue inflammation in a TRPV1 KO mouse were reported, endotoxin-induced airway inflammation41 or inflammation in the knee joint induced by capsaicin was attenuated by TRPV1 gene loss.42 Either sulfate induced colitis in mice or TRPV1 activation by dextran-enhanced neutrophil accumulation and histopathologic changes.43,44
The paradoxical role of the transient receptor potential vanilloid 1 receptor in inflammation
2010, Pharmacology and TherapeuticsTumour necrosis factor α mediates transient receptor potential vanilloid 1-dependent bilateral thermal hyperalgesia with distinct peripheral roles of interleukin-1β, protein kinase C and cyclooxygenase-2 signalling
2009, PainCitation Excerpt :This confirmed a bilateral hyperalgesia in the second mouse strain. TRPV1−/− mice had significantly higher baseline p.w.l. than WT mice (Fig. 2A, p < 0.05) as observed previously [3,26]. Furthermore, there was no difference from the baseline levels in the thermal hyperalgesic thresholds in the TRPV1−/− mice in either the Tyrode or the TNFα-treated paw at 4 h (Fig. 2A, p > 0.05 compared to the baseline value in the same paw), indicating that TNFα-induced thermal hyperalgesia is TRPV1 receptor-dependent.
Vanilloid receptor TRPV1-mediated phosphorylation of ERK in murine adjuvant arthritis
2009, Osteoarthritis and CartilageCitation Excerpt :The severity of adjuvant-induced arthritis (AIA) of the knee18,19 and ankle20 was found to be significantly reduced in TRPV1-KO mice. Also, intraarticular injections of capsaicin demonstrated significant attenuation of the articular vasoconstriction and reduction of blood flow in the knee of KO mice, indicating that these effects are TRPV1-dependent21. Although TRPV1 has been implicated in arthritis, the mechanisms are not fully understood.
