2018, Biomedicine and PharmacotherapyCitation Excerpt :We used enhanced chemiluminescence (ECL) reagents to detect immunoreactive bands. We cloned the human COX-2 promoter (a region spanning ―459 to +9 bp) [14–16] into pGL3-basic vector (Promega, Madison, WI, USA) to construct a COX-2-luc plasmid. The COX-2 luciferase promoter assay was performed as we have described [10].
Inhibition of nuclear factor-kappaB, cyclooxygenase-2, and metalloproteinase-9 expression by flavanols from evening primrose (Oenothera paradoxa) in human colon cancer SW-480 cells
2017, Journal of Functional FoodsCitation Excerpt :COX-2 has also been associated with CRC metastasis by stimulating the expression of MMP-9, which plays a key role in the degradation of the extracellular matrix (ECM) (Tsujii, Kawano, & DuBois, 1997). It is also known that promoter regions of both COX-2 and MMP-9 contain NF-κB-binding sites (Tazawa, Xu, Wu, & Wang, 1994). Consequently, over-expression of COX-2 and MMP-9, which can enhance malignant potential in CRC, may result from NF-κB activation.
