2018, Methods in EnzymologyCitation Excerpt :In all cases, incorporating hydrophobic general anesthetics into single-component membranes lowers the main chain transition temperature (TM) and disorders lipid chains (makes their motions more isotropic) at fixed temperature (Heimburg & Jackson, 2007; Kamaya, Ueda, Moore, & Eyring, 1979; Lorincz, Mihaly, Nemeth, Wacha, & Bota, 2015; Miller, Firestone, Alifimoff, & Streicher, 1989; Shieh, Ueda, Lin, & Eyring, 1976). These molecules also can impact the lateral pressure profiles, bending rigidity, and water permeability of simple membranes (Gruner & Shyamsunder, 1991; Jorgensen, Ipsen, Mouritsen, Bennett, & Zuckermann, 1991; Jorgensen, Ipsen, Mouritsen, & Zuckermann, 1993; Mountcastle, Biltonen, & Halsey, 1978; Trudell, Payan, Chin, & Cohen, 1975). Over the years, it has been argued that these types of physical effects are unlikely to contribute to the molecular mechanisms of general anesthetics, as they tend to produce small perturbations at physiologically relevant concentrations (e.g., ΔTM < 0.5°C) (Franks & Lieb, 1982, 1986, 1990).
