Group III receptors are expressed throughout the brain and are consistently found at presynaptic sites, enriched at the active zone (AZ) of glutamatergic synapses (Bradley et al., 1996, 1998; Corti et al., 2002; Kinoshita et al., 1996; Shigemoto et al., 1996, 1997). An exception is mGluR6 which is exclusively expressed by ON-bipolar cells in the retina and is enriched at dendritic tips contacting photoreceptors (Nomura et al., 1994).

2021, Neuroscience and Biobehavioral ReviewsCitation Excerpt :mGluR3 is widely expressed in the CNS, especially in the olfactory tubercle, dentate gyrus, cerebral cortex, lateral septal nucleus, striatum, substantia nigra pars reticulata, nucleus accumbens, amygdaloid nuclei, and cerebellar cortex (Tamaru et al., 2001; Tanabe et al., 1993; Testa et al., 1994). mGluR4 is mostly expressed in the cerebellum and also expressed in the olfactory bulb, cerebral cortex, hippocampus, lateral septum, striatum, thalamic nuclei, and pontine nuclei (Corti et al., 2002; Kinoshita et al., 1996; Makoff et al., 1996). mGluR7 is most extensively found in the whole brain, while mGluR6 is mainly expressed in the retina (Kinoshita et al., 1998; Nakajima et al., 1993).

Within the cerebellum, mGlu4 receptors have been reported to be present predominantly on the parallel fiber terminals (Kinoshita et al., 1996), in the molecular layer of the cerebellar cortex (Corti et al., 2002), where they reduce glutamate release and excitatory potentials at the PF-PC synapse (Abitbol et al., 2008; Antflick and Hampson, 2012; Goudet et al., 2012). Moreover, these receptors are present on inhibitory interneurons (Golgi, stellate/basket neurons, Berthele et al., 1999; Corti et al., 2002).
