Most current drug therapeutic approaches to Alzheimer's dementia are aimed either at elevation of the transient levels of acetylcholine in the brain or at direct enhancement of nicotinic receptor activation by the application of cholinesterase inhibitors or nicotinic agonists, respectively. One problem of the latter approach is agonist-induced desensitization of nicotinic receptors. An alternative option, which could circumvent this problem, involves a novel class of nicotinic ligands, which potentiate the response of nicotinic receptors to acetylcholine by acting from an allosteric site. The mechanism of action and the structure-function relationship of these potential drugs is discussed.

Challenges in designing therapeutic agents for treating alzheimer's disease-from serendipity to rationality

The process of rational drug designing together with serendipity has played an important role in the search for new drugs, for example, in neurotherapeutics the dopaminergic dysfunction of Parkinson’s disease, the dopaminergic hyperfunction of schizophrenia and the acetylcholine deficit in patients suffering from Alzheimer’s disease (AD). AD is the most common form of dementia which is a group of disorders that impairs mental functioning. The aim of this chapter is 1) to analyze some examples of discoveries in AD related disorders based on some serendipitous hypotheses and rational drug design approach such as structure based drug designing, ligand based drug designing and multi-target-directed ligand approach ii) to study the drug targets such as AChE, BACE-1, Tau protein for treating AD iii) to design second-generation agents with improved efficacy and safety iv) to illustrate the complexity of problems that have to be overcome for successful targeting and v) to study drug delivery and intercellular characterization of the blood brain barrier.
Pharmacological aspects of the acetylcholinesterase inhibitor galantamine

Several lines of evidence suggest that cholinergic deficits may contribute to the pathophysiology of psychiatric disorders as well as Alzheimer’s disease. There is growing clinical evidence that galantamine, currently used for the treatment of Alzheimer’s disease, may improve cognitive dysfunction and psychiatric illness in schizophrenia, major depression, bipolar disorder, and alcohol abuse. Since galantamine is a rather weak acetylcholinesterase inhibitor, but has additional allosteric potentiating effects at nicotinic receptors, it affects not only cholinergic transmission but also other neurotransmitter systems such as monoamines, glutamate, and γ-aminobutyric acid (GABA) through its allosteric mechanism. It is likely that these effects may result in more beneficial effects. To understand the underlying mechanism for the clinical effectiveness of galantamine, neuropharmacological studies have been performed in animal models of several psychiatric disorders. These studies suggest that not only the nicotinic receptor–modulating properties but also the muscarinic receptor activation contribute to the antipsychotic effect and improvement of cognitive dysfunction by galantamine. This review summaries the current status on the pharmacology of galantamine, focusing on its effect on neurotransmitter release and pharmacological studies in animal models of psychiatric disorders.
First asymmetric synthesis of (un)substituted bridged tetrahydro-2-benzazepines

A new and flexible route for the asymmetric synthesis of a variety of alkylated bridged tetrahydro-2-benzazepines has been developed. The key steps are the highly diastereoselective Michael addition of metalated SAMP-hydrazones to α,β-unsaturated esters combined with cyclomethylenation/Mitsunobu coupling reactions to secure the formation of the seven-membered azaheterocycle and of the bridged unit, respectively.
