Microwave accelerated Gewald reaction: synthesis of 2-aminothiophenes

Abstract

Microwave-promoted synthesis of 2-aminothiophenes by multicomponent reactions of a ketone with an active nitrile and elemental sulfur under KF-alumina catalysis is described.

    Many derivatives based on 2-amino/benzothiophenes were tested in mammalian carcinoma cells such as; hepatocellular (HEPG-2), pancreatic (MiaPaCa-2), cervical (HeLa), breast (MCF-7), colon (SW 620), laryngeal (HEP-2), and diploid fibroblasts (WI 38) with certain anti-proliferative effect [13–15]. They were also considered as milestone synthons in organic synthesis to produce many useful heterocyclic systems [16–18]. Gewald synthesis method with its various versions are the most appropriate synthetic methodologies to prepare the thiophene systems [19].
